Coumarins are widely distributed in a variety of natural sources, including plants. Coumarins are a subset of the larger group of compounds known as benzopyrones. The wide range of biological actions of coumarin derivatives, both natural and synthetic, have made them a topic of research. Its adaptability as scaffolds is largely responsible for their broad spectrum of pharmacological effects, including anticancer, anticoagulant, anti-inflammatory, anti-microbial, anti-HIV, and anti- Alzheimer's properties. Many compounds with the coumarin ring system have been synthesized by a variety of synthetic techniques. The project was carried out by synthesizing coumarin using the Pechmann Condensation method, and further Mannich reaction was carried out to prepare its 8-substituted (7-Hydroxy-4-methylcoumarin) derivatives. Nuclear magnetic resonance (NMR), infrared radiation (IR), and chromatography were used to characterize the derivatives (8-substituted,7- Hydroxy-4-methylcoumarin). Further in vitro research like antioxidant, anti-inflammatory, and anticoagulant activities on coumarin derivatives was conducted. The results showed us that listed compounds have antioxidant, anti-inflammatory, and anticoagulant properties. For the formulation process, compounds with strong antioxidant and anti-coagulant action were preferred. An anti-inflammatory gel formulation was developed, and its standardization and optimisation process were carried out.
